A child inherits half his/her DNA from the biological father and mother. As such, if the mother or father is not available for testing, the grandparents of the missing side can be used instead. Since only around 25% of a child’s DNA is inherited from each grandparent, both grandparents might be needed or the remaining mother or father should be included.
An grandparent test is more complicated than a normal paternity test because more tests, such as autosomal STR, Y-STR, and or mitochondrial DNA must be performed. Regardless of the number of tests, usually a buccal swab is used, which is a standard method for collecting DNA that is safe and quick. Blood samples, hair roots from plucked hairs, and other biological samples may also be used. The samples are purified for DNA, and the short tandem repeats (STR), sections of DNA that is different from person to person, is amplified using a technique called polymerase chain reaction (PCR). An expert then interprets the results and issues a report. This standard method is used worldwide and must be performed by trained, certified technicians.

Hair root
Collecting plucked hairs (with roots) is another option for DNA testing that is quick, easy, and almost pain-free. It is important to use plucked hairs as the DNA is mainly in the root part.
Blood spot
Collecting blood spots is an alternative that poses very little risk when a special device is used. Care must be taken during collection and the spots must be dried before posting the samples. DNA in the white blood cells is used for relationship testing.
Other biological evidence
If the above samples cannot be collected, other personal items can be used as a source of DNA, such as toothbrush or mask. However, this involves a slightly more complicated protocol and poses risk of contamination of DNA from other people.
